Cathepsin L of Sphaerospora molnari - localisation, function and diagnostic tools
NECHVILE, Rudolf Lukas
The aim of this thesis was to develop diagnostic and quantitative assays/tools based on western blotting, confocal microscopy and immunogold electron microscopy, as well as flow cytometry, to determine the localisation and potential function of the highly expressed cathepsin L of the myxozoan parasite Sphaerospora molnari (SmolCL) in its fish host, the common carp. Furthermore, a recombinant SmolCL was used in a vaccine trial to estimate its potential for raising antibodies in carp and test their immunoprotective potential towards S. molnari. This thesis provides new methodological tools for research and allows a greater understanding of myxozoan parasite-fish host interactions based on proteolytic enzymes.
Rozvoj zooplanktonu v rybnících s produkcí rychlených stádií hospodářsky významných druhů ryb
PECHA, Oldřich
This master thesis was focused on zooplankton development in ponds with advanced fry rearing of economically important fish species. For the purpose of my study, we sampled pond cascade that is managed by Štičí líheň - ESOX, spol. s r.o. in the village Liderovice, near Tábor. In this particular pond cascade, the annual production of advanced fry of economically important fish species takes place, namely pike, pikeperch, carp, tench and grass carp. Importantly, pond cascade is used several times during one growing season, and zooplankton is the main food source of all early stages of rared fish. For the production of advanced fry of fish served 3 ponds, which were released and refilled 2-3 times a season. Fourth pond served as a control stocked with one year old carp throughout the whole season. The water from an upper pond in the cascade served as zooplankton inoculum to a lower pond after the pond harvesting. Statistical analyses showed that the fry of different fish species in ponds lying in the same cascade does not affect the composition of zooplankton after restocking and water refilling. The only difference we noticed was in the treatment with pikeperch, which partially does not share the food base of other monitored species.We also found out, that the occurrence of Cyclops vicinus, Daphnia magna, D. ambigua or D. pulicaria was positively correlated with increased P-PO4 concentrations. We also confirmed that pond ecosystem can effectively retain nutrients from basins and use them for its production. As was shown after a short rain when the measured concentration of total phosphorus in the inflow in our pond system was 0.50 mg/L, on the contrary in the last pond of our cascade we measured values below 0.08 mg/L. High transparency was maintained in the whole cascade throughout the monitoring period, and an adequate economic profit was generated. Such management can be applied to only a few ponds (systems).
The possibilities of influence sensoric characteristics of common carp flesh
MAŠÍN, Milan
Organoleptic characteristics of fish meat are one of criteria, that make higher quality prerequisites for successful selling, especially for more exacting customers. An objective of this work is evaluate temperature and storage time aspects on these characteristics. Like an appropriate fish was selected Carp (Cyprinus carpio), weighing around 1,5 {--} 3 kg. In own sauce roast meat samples were sensoric analysed directly after killing and than in different time periods. The storage temperature was 3; 5 and -18 °C. Panel of evaluators judged odour, taste, flavour and consistence of fish meat. A protocol with an unstructured graphic scale was used for expressing the intensity of perception. Accomplishments were graphic and statistic processed.
Diet of great cormorant (Phalacrocorax carbo) in conflict localities
Ráslová, Pavlína ; Čech, Martin (advisor) ; Vrána, Pavel (referee)
vi Abstract Continental subspecies of the great cormorant (Phalacrocorax carbo sinensis) is the Czech Republic's most important fish-eating predator. It causes damage to production fish, as well as sport fishermen and even after its removal from the Czech list of endangered species in 2013, it did not disappear from the spotlight of both local fishermen and researchers. This thesis evaluates the qualitative and quantitative composition of the cormorant's diet during winter 2011/2012 at two selected locations: the Žehuň pond (Kolín district) and Berounka river in the natural reserve Zábělá near Pilsen. This work studies the main components of cormorant's diet in the selected locations and evaluates potential fluctuations and changes in its composition during winter. The study also conducted the analysis of the proportion of commercially harvested valuable fish in cormorant's diet and studied its diversity of the species spectrum. The resulting data for the two sites were compared with each other and with relevant literature. Regurgitated pellets analysis, as well as analysis of diagnostic bones and fish remains located at the sleeping roosts (night roosts) were used for the diet analysis. It was found that at both sites roach was the most caught fish, but from the point of view of total biomass, it was...

The study evaluates the state and development of fish stock in relation to the current fishery management and develops proposals for further management of the fish community and fishery regulations at the Lipno reservoir.
Haematological response of common carp \kur{Cyprinus carpio} to experimental infection with blood stages of the myxozoan parasite \kur{Sphaerospora molnari}
TOMANOVÁ, Anna
Sphaerospora molnari is a histozoic endoparasite causing severe inflammation during sporeformation in the gills of common carp. As for other myxozoans, little is known about the host response to early proliferation and multiplication of this parasite in the fish host. This thesis is focused on a controlled infection experiment using S. molnari blood stages and on determining cellular and haematological changes of common carp as a response to infection, over a 9-week period post intraperitoneal injection. The results show a noticeable increase in cellular components of innate immunity 14 dpi and a massive increase in lymphocyte numbers, likely B cells, 21-28 dpi. The data are discussed in relation to parasite infection intensity in various tissues and organs, and they strongly point to the acquisition of specific immunity of common carp to S. molnari resulting in a controlled, low level infection from 49 dpi onwards.
Kvalita prostředí a potravní chování kapra ve vztahu k přikrmování v polointenzivní rybniční akvakultuře
Adámek, Zdeněk ; Jurajda, Pavel ; Zukal, Jan ; Janáč, Michal ; Mikl, Libor ; Šlapanský, Luděk ; Všetičková, Lucie ; Prášek, Václav ; Šútovský, I. ; Chytka, V. ; Mrkvová, Markéta
Kromě menší hmotnosti mají i nižší obsah tuku.
